New Lumos has 8 Districts that you can complete in any order, but it's recommended to progress through the region in ascending order, numerically, as the enemies get increasingly harder to defeat.

Use Vampire and Elf

Vampire	High Stats and Tank Role Main Hero: ★★☆ Companion: ★★★ Best Personality: Energetic/Kind/Stubborn Unlockable after completing the Manor Macabre quest.
Elf	Support/Offensive Hybrid Main Hero: ★★★ Companion: ★★☆ Best Personality: Cool/Energetic Unlockable after defeating the Replica Dark Lord in Uncharted Galados (Galados Isle).

The Vampire and Elf are highly recommended to use when clearing areas in New Lumos and its succeeding regions. Vampires in particular will learn skills that can be useful for grinding your party, so once you've completed the Main Story, make sure to make it a top priority to unlock the Vampire.

Prepare for the Tower of Dread and Tower of Despair

Available after clearing New Lumos, the Tower of Dread and Tower of Despair are the most challenging areas in Miitopia. You'll need to make sure you have high-level equipment and eaten enough food to raise your characters' stats. With that, use your time in New Lumos to raise your party's levels and equipment.
