Yes, but It’s Not an Unlockable Weapon

Head to the Cemetery
Unlike in the original game, the Great Knife makes its appearance as an interactable object in the Labyrinth. It replaces the Rotating Cube in the cemetery and will be dropped as soon as you cross the Strange Wall.
