One of the best and quickest stages to farm Poes is Chapter 2: Growing Suspicion, with the presence of multiple Construct-type enemies and Shades in the Battle Zones of this level, as well as an extra section that features even more Shades to farm Poes for.
You can get around 500-800 Poes in this level, and you can even be faster when you use Rauru later when he has leveled up significantly, as he is the only playable character in this stage.

The best stage to farm Poes in the post-game is the In Search of Poes challenge. It is a Lv. 81-90 challenge added in the first free DLC update for the game. You can earn over 2 thousand Poes in a playthrough of this mission. However, this unlocks very late in the game when you may not have need for a lot of Poes anymore.
In Search of Poes Rewards and Materials

You will be able to gain Poe thru Combat in Battle Stages while in the Depths, the underground section of the map that unlocks during Chapter 1. The stages in the Depths contain several Constructs and Shades that you can farm poes from!

One of the most consistent enemy types to look for when hunting for Poes are Shades, ethereal red creatures that appear in stages. Hunting them down guarantees dropped Poes so locate them primarily in Depths stages and some Battle missions.

Another source of Poes in the game is defeating constructs, usually found in the Depths, in Challenge missions or Battle Quests. These constructs also emit the same red aura as the Shades, and will drop Poes upon defeat.

The main use of Poes is to buy items in the Talon & Tusk shop, which sells items which materials, some of which are affected by the Materials Sensor and Quest Sensors. Make sure to visit this shop when looking at Camps and Depots and get items you need!

Tusk & Talon is only available after completing the Camp Quest My Prized Treasures, as one of the camp upgrade quests.
Completing the Breaking into a New Market quest will allow Tusk & Talon to have a chance to appear in Supply Depots on the Hyrule Map, so that you won't need to enter a battle and find a camp in order to shop there.
Due to the Materials Sensor and Quest Sensor affecting what items may appear on the shop, we recommend you to buy the items you have tracked in the sensor, shortening farming time for these specific materials you are looking for.
As a note, Monster Essence is one of the mainstay items in the shop, and will always appear when the shop's items refresh.
