After transforming into Radiant Peach, the boss fight with Madame Grape will begin. The concept is pretty simple: you need to shoot Sparkle Blasts to damage Madame Grape who is standing in the middle of the stage.
Simply press B to shoot projectiles and aim towards Madame Grape's body. As the big bad villain takes hits, her feathers behind her body will disappear. Keep shooting until all the feathers are gone.
Madame Grape will protect herself by surrounding her with Sour Bunch Ghosts. They will block your line of sight and you won't be able to hit Madame Grape until they disappear, so attack them to clear up the room.
You can also reposition to a spot where there are no ghosts blocking the way. This is useful if you are attempting to clear the post game boss challenge for Madame Grape which requires you to avoid knocking out any ghosts.
Sparklers will constantly ignite and light up below the stage while you're shooting at Madame Grape. Keep moving to the left or right to avoid getting damaged.
Madame Grape will summon large presents that will fall from above. Move sideways and weave through the falling boxes to avoid getting hit.
Midway through the first phase, Madame Grape will summon a train that will run along the bottom of the stage. Fly upwards to avoid getting hit by the train.
If you're having a difficult time beating this boss, you can use the Heart Charm to get three extra hearts. You can turn this on in Sparkle Theater 1F and talking to the Theet near the center of the lobby.
Do note that there are no penalties when using a Heart Charm, so feel free to get extra hearts whenever you want the fight to be easier. You can always turn it off by speaking to the Theet again.
After defeating Madame Grape in Phase 1, a short cutscene will play where you will witness the boss transform into a fearsome giant and destroy the Sparkle Theater. Every Theet in the area will then grant their power to Peach and Stella, resulting to the princess transforming into Super Radiant Peach and starting Phase 2 of the boss fight!
As Super Radiant Peach, hold the B button to fire off your Sparkle Beam. Make sure to aim this attack towards Grape the Great's golden masquerade mask, which is her only weak point. You need to keep damaging her mask until it cracks and breaks.
While blasting away, you also need to dodge Grape the Great's projectile attacks, which she will constantly launch towards Peach throughout the fight.
Do note that Peach's Sparkle Beam attack will not clear any of the projectiles, and you will get hit if you try to direct a beam towards a projectile and fail to dodge in time.
Halfway through Phase 2, Grape the Great will spew out lasers from above.
The laser attack will be directed to the ground before it rises to where Peach is in the sky, so you can easily telegraph where the safe spots are to avoid getting hit.

After defeating Madame Boss, you'll be able to purchase Grape-themed Outfits for Peach and Stella at the Lobby Shop found in 1F. These cost 500 Coins.
You'll also be able to purcahse Radiant Outfits after using Radiant Peach during the boss fight. Both of these also cost 500 Coins.
After defeating Madame Grape, you'll also be able to check out the model for both Radiant Peach and Super Radiant Peach transformations in the Ensemble found in the Collections menu of the pause screen.
How to Unlock Radiant Peach Transformation
As the final boss of the story, defeating Madame Grape will grant you access to several post game content. These include Sparkle Decorations, boss challenges for all five Darkle bosses, and new collectibles in the form of locating Theets hiding within every level of the game.
After you finish the game, you can rebattle Madame Grape at the basement. There are three challenges you can attempt during the rematch, and you can get Sparkle Gems upon clearing them.
You first need to complete all levels on the fifth floor of the Sparkle Theater lobby before you can battle Madame Grape. Doing so will unlock all the third Peach Transformation levels found in the basement of the theater, because at this point, you will have completed the first and second stages per Transformation which are found from 1F to 5F.
Once you clear all four levels in 5F, a door will not appear in the middle of the fifth floor's lobby, so make you way down to the basement.
All Sparkle Theater Second Floor (2F) Levels
The next requirement needed to fight Madame Grape is completing all 10 levels found in the basement of the Sparkle Theater, which are the third levels for each Peach Transformation.
These levels will have you visit otherworldly versions of all the levels you've played in order to free the Sparkla Theets trapped within.
All Sparkle Theater Basement (BF) Levels
Like with the previous boss fights, you also need to give Stella Sparkle Gems before you can begin the boss battle. For the Madame Grape fight, you need 30 Sparkle Gems to open the boss door.
The cutscene will show that it won't be enough to unlock the door, but fret not, as all the Sparklas you've saved in the basement levels will lend you their power!
