The Platinum Trophy for Hell is Us is has some missable trophies and out of the way trophies which require exploration and backtracking which will take a bit of time. All trophies can be obtained on any difficulty.
You can acquire most of the trophies on your first playthrough of the campaign, provided that you take the extra effort in collecting all research items but you'll most likely need to backtrack for any Mysteries and Good Deeds you might've missed. Here's a step-by-step guide on getting all the achievements and trophies in the game:
1. Complete The Main Story

Progressing and finish the main story allows you to unlock all of the locations you can travel to in Hell is Us. This rewards you with most of the trophies for the Platinum, including the trophies related to enemies since you'll be fighting most of them throughout the story.

2. Upgrade Your Weapons and Defensive Gear
The loadout of the game allows you to wield two weapons and two defensive gear. Focusing your upgrades and leveling up on these equipment will help you get through the story faster and make enemy encounters easier.
Later on in the game, you can enhance one weapon for other types you didn't enhance to obtain one of the trophies. Having the right glyphs and relics also matter, so make sure to equip them.

You can get 21 trophies just by completing the main story itself and fighting most of the enemies you encounter. This is considering that you do a single Mystery and Good Deed, which shouldn't be hard since at least one of them are part of the story.
3. Complete All Mysteries and Good Deeds

The other trophies are related to both Mysteries and Good Deeds. Mysteries are puzzles that are triggered from collecting research items and notes which leads to treasures and items. Good Deeds are similar but the difference is they allow you to help NPCs throughout Hadea.
Getting the trophies for both of them will require you to backtrack to previous locations or even do it on another playthrough if you've missed them. You'll need to accomplish all of them to get the trophies.

4. Acquire All Relics and Glyphs
Some trophies also require you to obtain all of the relics and glyphs. This can take some time as well since you get some of them from Mysteries and Good Deeds, while you get others from the vaults scattered throughout Hadea.
Getting these trophies requires you to backtrack to previous areas after finishing the game since you won't get all of them on your first playthrough if you don't go out of your way to get them.

5. Close All Timeloops and Defeat Their Guardians
A trophy is also tied to closing all of the Timeloops and defeating all of the Timeloop Guardians. You'll have to use the Guardian Detector that you get during the story do locate most of the Guardians.
You can also complete the story without closing all of the Timeloops, so you'll have to backtrack and close all of them to get the trophy.

6. Get All Research Items
This trophy will probably take the longest in this part since it requires you to get all of the research items in the game. This can be time consuming since they're scattered throughout Hadea and will need you to explore every location thoroughly to get all of them.
