Known to friends and family as Peter Parker, it has been 8 years since he was bit by the radioactive spider that gave him his current powers and abilities. He has since been protecting New York from enemies as their friendly neighborhood Spider-Man.
Mary Jane Watson has been best friends with Peter ever since middle school. She works as a reporter for The Bugle where she grew a following for her articles. MJ and Peter maintain a professional relationship as she continues to help his investigations as Spider-Man.
Yuri Watanabe is the Captain of the New York Police. While she currently works in a more managerial role, she is also very skilled in hand-to-hand combat. Spider-Man works very closely with her, often receiving first-hand intel on crimes in the city.
As a scientist with genius-level intellect, Otto Octavius is able to devise elegant and effective solutions to even the most complex problems. While other kids would have superheroes like Captain America or Iron Man as their childhood idol, Peter always looked up to him.
After leaving Oscorp, the company he co-founded, he founded and currently works at his own company, Octavius Industries. Peter was initially hired for a summer internship but has since been working full time for him.
Together with her late husband Ben, May Parker took in and raised Peter after the death of his parents. Her kind and generous spirit often inspire the people at F.E.A.S.T., where she works as the Operations Manager.
After being orphaned at the tender age of seven, Martin Li grew up in foster care in Manhattan. At the age of 18, he went to China and became a billionaire in the shipping industry. Nine years later, he returned to New York and put up the F.E.A.S.T. homeless shelter.
Norman Osborn is the co-founder and CEO of Oscorp, the world's biggest tech company. He is also the current mayor of New York where he often uses his political power to carry out his agenda. Still, Peter doesn't harbor any ill will towards him since he raised his best friend, Harry.
A veteran of the police force, Jefferson Davis is one of the few officers to treat Spider-Man as an equal rather than a nuisance. His time on the force has made him highly skilled and also very knowledgeable on organized crime in the city, particularly on Wilson Fisk's operations.
Miles Morales is the son of police officer Jefferson Davis. He is currently a student with an intuitive grasp of science and technology. After the death of his father, Peter takes it upon himself to help Miles in any way.
After the death of her husband, Jefferson, Rio remains a source of positivity to those around her. She continues to take care of her son, Miles and takes the time to make sure Peter is also alright.
Born into royalty as the daughter of Ernst, king of the Eastern European nation of Symkaria, Silver Sablinova is the CEO of paramilitary force Sable International. After police officers on Wilson Fisk's payroll were arrested, Mayor Osborn hired her company to reinforce the dwindled police force.
Dr. Morgan Michaels is the top researcher for GR-27 at Oscorp. His specialization in hereditary genetic diseases and finding cures for them has made him an extremely gifted biochemist. Being the current project lead for GR-27, also known as Devil's Breath, his understanding on the serum is unrivaled.
Wilson Fisk is one of New York's most powerful criminals. He controls the police, judges, and the city council, among other things. Years of training his agility and dexterity, coupled with his mental fortitude has made him one of Spider-Man's most formidable foes.
With the use of vibro-shock units of his own design, Herman Schultz aka Shocker was one of the first super
adversaries Spider-Man faced. This encounter would prove fruitful for Spider-Man as he would develop a sense of concern for civilians.
If he assessed that engaging in fight would harm any innocent bystanders, he would make sure to back off and catch them at a more opportune moment.
With his near invincibility and superhuman strength, Tombstone is a one of Spider-Man's more ruthless enemies. He primarily relies on his strength when fighting rather than outsmarting his opponent which would often result in losses against Spider-Man. That being said, he relishes a good challenge and always looks forward to the next fight.
Mac Gargan used to be an average street criminal until he signed up to fuse his body with a mechanical suit and be known as Scorpion. His mechanical suit gives him enhanced strength, speed, endurance, and agility along with the ability to scale walls with his claws while his tail can be used a blunt weapon.
An absolute unit of a man, Aleksei Sytsevich was an enforcer for the Mob before acquiring an experimental combat suit and becoming known as the Rhino. His extreme strength and resistance to extreme conditions combine for a very strong and durable opponent. Even with Spider-Man's accelerated healing, it took him weeks to be able to recover from their first encounter.
After a freak accident that gave him the ability to control electricity, Max Dillon began his criminal career as Electro. He was still getting used to controlling his powers when he first fought Spider-Man and subsequently burned himself out. Spider-Man speculates that if Electro ever did tap into the full potential of his powers, not even the Avengers could stop him.
At the age of 76, Adrian Toomes may be the oldest adversary of Spider-Man. With his background as a former electronics engineer, he was able to make a flight suit that gave him enhanced strength and the ability to fly at speeds of up to 95 mph. It took a few years before Spider-Man could defeat The Vulture and even then he proved to be tricky opponent.
As the alter ego of Martin Li, Mister Negative has the ability to generate and control negative energy. He can effectively control people's minds and even give a small portion of his abilities to members of his terrorist group, the Inner Demons.
After ignoring Peter's warning about the effects of the neural interface on his mind, Otto Octavius turned into the revenge-obsessed Doctor Octopus. With his new mechanical arms giving him superhuman strength and the ability to scale almost any surface, he is hellbent on exacting his revenge on his former partner, Norman Osborn.
Even with all the malevolent deeds Doctor Octopus has done, Peter still believes that Dr. Octavius can be saved.
With the ability to mimic moves and the powers of his enemies, Taskmaster has arrived in New York to study Spider-Man. He has set up multiple "challenges" across the city to learn how Spider-Man operates and how to replicate his powers. While his motives behind these challenges are unclear, Spider-Man is ready to fight him at his own game.
While the longtime green-skinned villain isn't in the game, Norman Osborn (the first incarnation of Green Goblin) appears in the game in a different capacity — the Mayor of New York.

Venom does not appear in the game but a post-credits scene suggests he might already found a host.
