You can not lock on to targets during combat in Ghost of Tsushima. The only way to target is to to direct your attack to a nearby enemy. This makes the combat difficult at times, due to not being able to accurately attack the intended target and the camera ending up in an awkward position, blocking your view.
You will need to keep in mind your camera position during combat and adjust accordingly while fighting. This can be tought, but can be made easier by increasing the camera speed in the Gameplay Option in the Pause Menu.
In melee combat there is no way to lock on to targets, but for ranged there is some thing close enough. Turning the Aim Assist on will allow you to automatically target to the closest enemy to your crosshair with a ranged weapon.
